You can only adjust the gain if using the front two line inputs (Inputs 1 and 2). The rear two (Inputs 3 and 4) have a fixed gain level applied to them. The level can be altered using Focusrite Control 2 if listening live, or in your DAW if recording, but the gain can not be changed.
