Mine came with a hypodermic needle.. is this for injecting lighter fluid?

Mine did not so not sure why it would have. You just dip it in alcohol, no injecting required.

What's the best thing to use to light the rope?

First of all, dip the wick of the stem or rope in isopropyl alcohol. That will ensure that the wick stays lit (and has the added benefit of being able to practice with one of the stems and actually re-use it). Then you can light it with a match or a normal cigarette (bic) lighter.
